I don't see any immediate blotting numbers after either 2/off 1/off or 6/4 5/4. In fact, after 2/off 1/off, I don't see any sequences of two rolls that leave a blot, whereas after 6/4 5/4 there are some (e.g., 6-3 6-3, or 6-6 5-2, maybe others). So I don't see any downside to taking two off.
If we move two of Black's checkers from the 2-point to the 1-point, then after 2/off 1/off, we have some sequences of two rolls that blot (e.g., 6-2 6-2 or 5-2 5-2 or 6-2 5-2 or 5-2 6-2). That looks like a closer decision.
